Sample types

CTD

CTD sampling ideally occurs 1x per day close to local noon/solar noon. If the CTD takes place within ~2 hours of local/solar noon, then this sampling effort can replace the 1200 inline sampling timepoint. Cruises that accommodate Bio-GO-SHIP sampling tend to have a separate CTD cast "bio-cast" once per day that goes to a max depth of 1000 m.

eDNA Depths (m): 1000, 200, 100, 5. x1 sterivex each depth

RNA Depths (m): 5. x1 sterivex each depth

HPLC Depths (m): 100, 40, 5. x1 GF/F each depth

POM Depths (m): 5. x3 GF/F for POC/N, x3 GF/F for POP

FCM Depths (m): 1000, 500, 200, 150, 100, 75, 40, 5. ~2 mL sample each depth

Transmissometer Continuously collecting data for all CTD casts (not just biocasts). Lenses must be cleaned with isopropyl and DI 1x per day. Lenses covered with electrical tape during CTD cast for a calibration "dark cast" ~1x per week.

Underwater Vision Profiler (UVP5) Managed by Bio team and PI Steffi O'Daly. Continuously collecting data for full depth CTD casts (settings are switched to not collect data for any shallow casts including Bio-casts).

Inline

Inline sampling ideally uses a diaphragm or positive-displacement pump (minimizes cellular damage caused by impeller pumps) and sample water is gathered at a sink located closest to the inlet valve of surface water (minimizes shear). Inline sampling occurs 3x per day for discrete sampling, and continuously for cytometry and optics.

Discrete timepoints (local/ship-time): 0600, 1200, 2000

Morning (0600): eDNA (x1 sterivex), HPLC (x1 GF/F), POM (x3 GF/F for POC/N, x3 GF/F for POP), FCM (x1 2 mL sample)

Noon (1200): eDNA (x1 sterivex), RNA (x1 sterivex), HPLC (x1 GF/F), POC/N small volume (x1 GF/F + blanks), POM (x3 GF/F for POC/N, x3 GF/F for POP), FCM (x1 2 mL sample)

Evening (2000): eDNA (x1 sterivex), HPLC (x1 GF/F), POM (x3 GF/F for POC/N, x3 GF/F for POP), FCM (x1 2 mL sample)

Midnight (0000): eDNA (x1 sterivex), RNA (x1 sterivex)

Large volume particulate organic matter (POC/N and POP) Volume: 9 L, triplicates of each
